چکیده

Rationale There is no optimal treatment available for symptomatic degenerative disc disease which affects millions of elderly people worldwide. Most commonly used treatment modality is only symptomatic pain relief by drugs. An alternative approach is the surgical correction of intervertebral disc (IVD). Engineering of in vitro intervertebral disc composites of predefined size and shape starting from appropriate scaffolds, combined with autologous cells, has the potential to overcome the limitations associated with typical treatments.

Oriented lamellar silk fibrous scaffolds to drive cartilage matrix orientation: towards annulus fibrosus tissue engineering.

A novel design of silk-based scaffold is developed using a custom-made winding machine, with fiber alignment resembling the anatomical criss-cross lamellar fibrous orientation features of the annulus fibrosus of the intervertebral disc. Crosslinking of silk fibroin fibers with chondroitin sulphate (CS) was introduced to impart superior biological functionality. The scaffolds, with or without CS...

Strategies for replicating anatomical cartilaginous tissue gradient in engineered intervertebral disc.

A critical challenge in fabricating a load bearing tissue, such as an intervertebral disc, is to simulate cellular and matrix alignment and anisotropy, as well as a specific biochemical gradient. Towards this goal, multilamellar silk fibroin scaffolds having criss-cross fibrous orientation were developed, where silk fibers in inner layers were crosslinked with bioactive molecule chondroitin sul...

Adipose Stem Cells and Intervertebral Disc Regeneration:

General Abstract Current treatments for the degenerated intervertebral disc (IVD) comprise pain management and invasive surgical interventions; both of them are aimed at relieving the pain instead of repairing the tissue and restoring the biological function of the disc. Cell-based tissue engineering may meet the clinical need for the repair or regeneration of a damaged tissue, and it might be ...

Comparing Behavior of Chondrocyte Cells on Different Polyhydroxybutyrate Scaffold Structure for Cartilage Tissue Engineering

Introduction: As the ability to repair cartilage tissue in body is limited, finding a suitable method for cartilage regeneration has gained the attention of many scholars. For this purpose, scaffold structure and morphology, along with cell culture on it, can be a novel method to treat cartilage injuries, osteoarthritis. Methods: In this study, polyhydroxybutyrate (PHB) is selected as the scaf...

A Structurally and Functionally Biomimetic Biphasic Scaffold for Intervertebral Disc Tissue Engineering

Tissue engineering offers high hopes for the treatment of intervertebral disc (IVD) degeneration. Whereas scaffolds of the disc nucleus and annulus have been extensively studied, a truly biomimetic and mechanically functional biphasic scaffold using naturally occurring extracellular matrix is yet to be developed. Here, a biphasic scaffold was fabricated with collagen and glycosaminoglycans (GAG...
